Update
Orforglipron is approved, and it is not a peptide. Both halves matter.
Approved on 1 April 2026 as Foundayo: the first GLP-1 that is a true small molecule, taken as a tablet with no food or timing rules. Across the 94 vendors we read, exactly one sells it — and the chemistry explains why.
The FDA approved orforglipron on 1 April 2026, under the brand name Foundayo, for chronic weight management in adults with obesity or with overweight and a weight-related condition. It is a tablet taken once a day, at any time, with or without food.
The part that gets flattened in most coverage is that orforglipron is not a peptide. Every other GLP-1 receptor agonist is a modified copy of the hormone, and peptides are digested, which is why the rest of the class is injected. Orforglipron is a small molecule designed from scratch to fit the same receptor, and small molecules survive the stomach.
That is also not the same claim as oral semaglutide. Rybelsus is a peptide carried past the stomach lining by an absorption enhancer, which is why it comes with instructions about an empty stomach, a specific small volume of water, and half an hour of waiting. Orforglipron has none of those, and the absence of the rule is the clinically interesting part rather than a marketing line.
The efficacy is real and it is smaller than the injections. In the ATTAIN-1 phase 3 trial, 3,127 adults with obesity or overweight with a comorbidity took 6, 12 or 36 mg once daily for 72 weeks and lost roughly 7.5% to 11.2% of body weight by dose, against 2.1% on placebo. Injected semaglutide and tirzepatide report larger mean reductions in their own trials. A pill that is easier to take and works somewhat less well is a genuine trade, not a free upgrade, and anyone choosing between them should see it as one.
Here is what our own data adds. Across the 94 vendors in our directory, we hold confirmed listings for semaglutide from 26, tirzepatide from 32 and retatrutide from 34. For orforglipron we hold one, and have done since we started tracking it. The research-chemical market has effectively not picked this compound up.
The chemistry is a reasonable explanation for that. The grey market for these drugs runs on peptide synthesis: buy the active ingredient in bulk, freeze-dry it into vials, ship a powder for the buyer to reconstitute. A small molecule is a different supply chain with different equipment and different intermediates, and the existing one does not transfer to it. Whether that holds as demand grows is an open question, and it is the kind we will be able to answer from the index rather than by guessing.
Two practical notes for readers of this site. Nothing about orforglipron is reconstituted, so the peptide calculator does not apply to it and its profile says so. And for once the approved product and the research-chemical version are not the same molecule in different packaging: people buy retatrutide because there is no approved version of retatrutide, whereas orforglipron has one, available on prescription, made to a pharmaceutical standard, with a regulator able to act if a batch is wrong.
