Storing peptides properly

Dry powder and mixed solution have completely different shelf lives. The clock that matters starts when you add water.

The single most useful thing to understand about peptide storage is that reconstitution changes everything. A freeze-dried vial is stable for a long time; the same peptide in solution is not. Most storage mistakes come from applying the first number to the second situation.

Before mixing

Lyophilised powder is stable because there is essentially no water for chemical degradation to happen in. Kept cold, dry and dark, an unopened vial lasts far longer than most people assume.

Refrigeration at 2–8 °C is the normal recommendation. For long-term holding, a freezer at −20 °C or colder extends that considerably, to years instead of months for many compounds, provided the vial stays sealed and dry.

Light and moisture are the two things to protect against. Vials arriving with a desiccant are telling you which one the manufacturer worried about.

After mixing

Once water is in the vial, refrigerate it and keep it refrigerated. Most compounds are treated as good for around four weeks at 2–8 °C, though this varies and the figure on each compound page is the one to follow.

Do not freeze a reconstituted vial. Ice crystal formation damages peptides, and repeated freeze-thaw cycles are worse still.

Bacteriostatic water is what makes repeated entry possible at all. The benzyl alcohol in it prevents bacterial growth. Plain sterile water does not, and a multi-use vial mixed with it is not safe to keep.

  • Dated label on every mixed vial

    Date and concentration, written when you mix it. This is the cheapest error prevention available.

  • Freezing a reconstituted vial

    Ice damages peptides in solution. If a mixed vial has frozen, discard it.

  • Leaving a mixed vial out overnight

    Bacteriostatic water slows bacterial growth; it does not make a vial shelf-stable.

Travel

Cold chain and documentation are the two things that go wrong. An insulated container with a cool pack handles the first. For the second, keep vials in their original labelled packaging and check the rules of wherever you are going before you leave rather than at the airport.

Do not put anything temperature-sensitive in checked luggage. Hold luggage gets hot, gets cold, and gets lost.

Terms used here

Peptide
A short chain of amino acids: the same building blocks as a protein, in a chain short enough to behave differently.
Sterile technique
The handling that keeps a vial and an injection free of contamination: clean hands, an alcohol-wiped stopper, a fresh needle every time.
Vial
The small sealed glass container a peptide is sold in, holding a stated mass of freeze-dried powder under a rubber stopper.
Cold chain
Keeping something refrigerated at every step from manufacture to use. Peptides ship without one far more often than the storage instructions suggest.
Reconstitution
Adding liquid to a freeze-dried peptide powder to turn it into a solution that can be drawn into a syringe.
Bacteriostatic water
Sterile water containing a small amount of benzyl alcohol, which stops bacteria multiplying and allows a vial to be used more than once.
Lyophilised
Freeze-dried. The process used to turn a peptide solution into a stable dry powder for shipping and storage.
